01
Adamax is a synthetic analogue of Semax. It keeps Semax’s Met-Glu-His-Phe-Pro-Gly-Pro sequence, adds an acetyl group to the N-terminus, and replaces the free C-terminus with an adamantane-based carboxamide group, the same terminal unit used in the CNTF-derived peptide P21 (P021).
The design aims to combine Semax’s core sequence with greater lipophilicity and resistance to enzymatic breakdown. Unlike Semax, Adamax itself has very little peer-reviewed research behind it, so most of its proposed properties are extrapolated from Semax and from the adamantane strategy used in P21. It is supplied as a lyophilized reference material for in vitro and analytical research only.
02
Adamax is Semax with both ends modified.
Semax core
Met-Glu-His-Phe-Pro-Gly-Pro: ACTH(4–7) followed by the Pro-Gly-Pro tail.
N-terminal acetyl group
Blocks aminopeptidases and removes the N-terminal positive charge, as in N-Acetyl Semax Amidate.
Adamantane group
Adamantane is a rigid, cage-shaped hydrocarbon (C₁₀). Here it is incorporated as a 3-aminoadamantane-1-carboxamide unit at the C-terminus. It is very hydrophobic and bulky, which increases lipophilicity and sterically hinders C-terminal enzymatic cleavage.
Overall properties
The molecule is markedly more hydrophobic and about 218 Da heavier than Semax. Met1 remains the main oxidation-sensitive site.
03
Because direct studies are scarce, research on Adamax is best framed as comparative.
Extrapolation from Semax
Its proposed effects on neurotrophic signalling are inferred from Semax research, which reported changes in BDNF and TrkB expression in animal models. These findings have not been established for Adamax itself.
Adamantane strategy
Adamantane groups are used in medicinal chemistry to increase lipophilicity and metabolic stability; P21 is a peptide example of the same approach.
Open questions
Whether the bulky C-terminal group changes receptor interactions or biological activity relative to Semax has not been well characterised and is best tested directly.
These points are research context only; no effects are claimed for this material.
04
Analogue series comparisons
Studied alongside Semax and N-Acetyl Semax Amidate to measure how increasingly modified C-termini affect stability, hydrophobicity and activity in vitro.
Stability assays
Used in serum and tissue homogenate incubations to compare degradation rates with its parent peptide.
Hydrophobic peptide analytics
A test compound for reversed-phase methods for peptides carrying bulky hydrophobic groups.
05
Storage
Store the lyophilized powder at −20 °C, sealed, desiccated and protected from light.
Oxidation
Met1 oxidises in the presence of oxygen, trace metals and light. Minimise air exposure.
Reconstitution
The adamantane group lowers water solubility compared with Semax. Water or dilute acetic acid usually works for moderate concentrations; a small amount of DMSO can help with concentrated stocks.
Solutions
Prepare fresh or freeze single-use aliquots, and use low-binding polypropylene tubes, as hydrophobic peptides adsorb to surfaces.
06
Analysis must confirm both terminal modifications.
HPLC
Reversed-phase C18 or C8 with a TFA gradient. Adamax retains much longer than Semax or N-Acetyl Semax Amidate because of the adamantane group. The Met-sulfoxide form elutes earlier.
Mass spectrometry
Positive-mode ESI shows [M+H]⁺ at m/z 1032.5 and [M+2H]²⁺ at m/z 516.8 (monoisotopic). MS/MS confirms the acetyl group and the adamantane-containing C-terminal fragment.
Impurity profile
Typical checks include Met1 oxidation (+16 Da), non-acetylated and incompletely coupled forms, truncated sequences and counter-ion content.
07
It has the same seven-residue Semax sequence, with an acetyl group on the N-terminus and an adamantane-based carboxamide group at the C-terminus.
It is a rigid, very hydrophobic hydrocarbon cage. It increases lipophilicity and sterically hinders enzymes that act on the C-terminus. The same terminal group is used in P21.
Very little on Adamax itself. Most of its proposed properties are extrapolated from Semax and from the adamantane strategy used in P21, so direct comparative testing is recommended.
No widely recognised CAS number has been assigned. Identity is confirmed by formula (C₅₀H₆₉N₁₁O₁₁S), accurate mass and MS/MS.
It retains much longer on reversed-phase columns because of the hydrophobic adamantane group, so gradients usually need a higher organic content.
No. It is supplied strictly as a reference compound for laboratory and analytical research and is not for human or animal use.
