Research Overview

Eloralintide (development code LY3841136) is a synthetic, long-acting analogue of human amylin. Like cagrilintide, it is built on the 37-residue amylin scaffold and carries a C20 fatty diacid for albumin binding, but it was engineered to favour one amylin receptor subtype, AMY1, over the calcitonin receptor.

Published cell assays report an EC₅₀ of about 24 pM at the human AMY1 receptor, compared with about 254 pM at AMY3 and about 291 pM at the calcitonin receptor, roughly a 12-fold preference for AMY1. This makes eloralintide a useful contrast to the non-selective amylin/calcitonin agonist cagrilintide. It has progressed from preclinical discovery into clinical studies. The material listed here is supplied as a lyophilized reference compound for in vitro amylin-receptor pharmacology and analytical work on lipidated peptides.

Structural & Sequence Context

Eloralintide keeps the overall design of amylin while changing features that control selectivity, stability and duration of action.

Amylin scaffold
The 37-residue chain retains amylin’s N-terminal ring and its receptor-binding region, with an amidated C-terminus.

Engineered substitutions
The published structure includes several non-standard residues in the amylin sequence. Together, these changes shift activity towards the AMY1 receptor and improve resistance to enzymes and to amyloid formation.

N-terminal ring
As in native amylin, residues 2 and 7 close a small ring that is needed for receptor activation. In the published structure, this ring is described as a carbon-bridged (methylene thioacetal) link rather than a disulfide, making it resistant to reducing agents. Confirm the bridge form against the certificate of analysis for each lot.

Lipidated Lys26
A C20 fatty diacid is attached to the side chain of Lys26 through a γ-glutamate-based linker. This provides albumin binding, as with cagrilintide and the incretin analogues in this category.

03

Mechanistic Research Context

Eloralintide is characterised in vitro by comparing its activity across the amylin and calcitonin receptors.

Receptor profile
Amylin receptors are the calcitonin receptor paired with RAMP1 (AMY1), RAMP2 (AMY2) or RAMP3 (AMY3). In cAMP assays in UM-UC-3 cells expressing the human receptors, eloralintide showed EC₅₀ values of about 24 pM (AMY1), 254 pM (AMY3) and 291 pM (calcitonin receptor).

Selectivity in context
The preference is for AMY1 rather than exclusive: eloralintide still activates AMY3 and the calcitonin receptor at higher concentrations. Cagrilintide, by contrast, is described as a non-selective amylin/calcitonin agonist.

Preclinical and clinical context
Published rat studies report reduced food intake and fat-mass-driven weight loss, and the compound has been evaluated in clinical studies. These findings are cited only as background.

Characterisation of the reference material itself is limited to in vitro and analytical work.

Laboratory Applications

Amylin-receptor selectivity panels
A reference AMY1-preferring agonist in cAMP and binding assays at AMY1, AMY2, AMY3 and the calcitonin receptor, compared with native amylin, pramlintide and cagrilintide.

Lipidated peptide HPLC
Its C20 diacid makes it a demanding test compound for reversed-phase methods for acylated peptides.

Ring-integrity methods
Used to compare how a carbon-bridged ring and a disulfide ring behave under reducing conditions in intact-mass and peptide-mapping methods.

Multi-target metabolic panels
Provides the selective amylin reference alongside semaglutide, tirzepatide, mazdutide, survodutide and retatrutide.

Stability & Handling

Storage
Store the lyophilized powder at −20 °C, sealed, desiccated and protected from light. Let the vial reach room temperature before opening.

Reconstitution
DMSO dissolves it well for concentrated stocks. Aqueous solubility depends on the salt form (for example sodium or acetate) and on pH, so check the certificate of analysis or test a small amount first. Avoid vigorous vortexing.

Ring stability
A carbon-bridged ring is not opened by reducing agents, unlike a disulfide. Even so, avoid harsh oxidising or strongly acidic conditions, as the ring is still essential for activity.

Aggregation
Amylin-derived peptides can self-associate at high concentration. Thioflavin-T fluorescence or dynamic light scattering can be used to check concentrated stocks.

Labware
Use low-binding polypropylene tubes and tips, and prepare dilutions shortly before use.

06

Analytical & Purity Considerations

Analysis must confirm the sequence, the N-terminal ring and the lipid side chain.

HPLC
Wide-pore (300 Å) C4 or C8 columns at elevated temperature usually give the best peak shape and recovery for this lipidated peptide. Use formic acid rather than TFA when coupling to MS.

Mass spectrometry
ESI-MS shows multiply charged ions that deconvolute to about 4526.1 Da. If the ring is carbon-bridged, the intact mass does not change after treatment with a reducing agent, which distinguishes it from disulfide-bridged amylin analogues such as cagrilintide.

Structure confirmation
Enzymatic digestion with LC-MS/MS confirms the ring-containing N-terminal fragment and the attachment of the lipid side chain to Lys26.

Impurity profile
Typical checks include the des-lipid peptide, deamidation of asparagine and glutamine residues (+1 Da), truncated sequences and the free-acid C-terminus.

Frequently Asked Questions

How does eloralintide differ from cagrilintide?

Both are lipidated amylin analogues, but eloralintide prefers the AMY1 receptor (about 12-fold over the calcitonin receptor in published assays), whereas cagrilintide activates amylin and calcitonin receptors non-selectively. Their sequences, lipid attachment sites and N-terminal ring chemistry also differ.

Why use a carbon bridge instead of a disulfide?

A disulfide can be opened by reducing agents or rearranged at basic pH. A carbon-bridged (thioacetal) ring, as described in the published structure, keeps the same ring shape needed for receptor activation while resisting reduction. Confirm the bridge form for each lot on the certificate of analysis.

How is eloralintide identified by mass spectrometry?

By its intact mass of about 4526.1 Da and by LC-MS/MS of digested fragments, which confirm the amylin-derived sequence, the N-terminal ring and the lipid on Lys26. A carbon-bridged ring also shows no mass change after reduction, unlike a disulfide.

What gives eloralintide its long-acting profile?

Mainly the C20 fatty diacid on Lys26, which binds serum albumin. The engineered substitutions and the reduction-resistant ring also improve stability.

What chromatographic conditions suit eloralintide?

Wide-pore (300 Å) C4 or C8 reversed-phase columns at elevated temperature, with formic acid in the mobile phase for LC-MS. The exact gradient and temperature are set during method development.

How selective is eloralintide?

Published cAMP data show EC₅₀ values of about 24 pM at AMY1, 254 pM at AMY3 and 291 pM at the calcitonin receptor. It prefers AMY1 by roughly 12-fold but still activates the other receptors at higher concentrations.

Is eloralintide intended for human or veterinary use?

No. It is supplied strictly as a reference compound for laboratory and analytical research and is not for human or animal use.
