Research Overview

N-Acetyl Epitalon Amidate is Epitalon (Ala-Glu-Asp-Gly) with both ends capped: an acetyl group on the N-terminal alanine and an amide in place of the C-terminal carboxylic acid. Its sequence is written Ac-Ala-Glu-Asp-Gly-NH₂.

Free peptide termini are the main targets of aminopeptidases and carboxypeptidases, so capping both ends is expected to make the peptide more resistant to enzymatic breakdown in biological samples. The core sequence is unchanged. It is supplied as a lyophilized reference material for in vitro research, particularly for side-by-side comparison with Epitalon in stability and analytical studies.

02

Structural & Sequence Context

The analogue keeps Epitalon’s four residues and modifies only the ends.

Core sequence
Ala-Glu-Asp-Gly, as in Epitalon.

N-terminal acetyl group
Replaces the free amine on Ala1 with an acetamide. This blocks aminopeptidases, removes one positive charge and adds 42 Da.

C-terminal amide
Replaces the carboxylic acid on Gly4 with a carboxamide. This blocks carboxypeptidases, removes one negative charge and lowers the mass by about 1 Da.

Charge
Because one positive and one negative terminal charge are both removed, the net charge stays at about −2, carried by the glutamate and aspartate side chains.

Retained weak point
The internal Asp3-Gly4 bond is unchanged, so the risk of succinimide and isoaspartate formation is the same as in Epitalon.

03

Mechanistic Research Context

Research interest in the capped form is mainly comparative.

Relation to Epitalon research
Published work on Epitalon includes studies of telomerase (hTERT) expression in cultured human cells, pineal and circadian gene expression, and age-related signalling. Whether the capped analogue reproduces these effects has not been established and is best tested directly.

Enzymatic stability
Blocking both termini is expected to slow breakdown by terminal peptidases, which can be measured by incubating both forms in serum or tissue preparations and tracking the intact peptide by LC-MS.

Assay windows
A more stable analogue can, in principle, allow longer in vitro incubations, but this must be confirmed in each assay system.

These points are research context only; no effects are claimed for this material.

04

Laboratory Applications

Capped versus uncapped comparisons
Paired with Epitalon to measure how terminal capping changes stability, chromatography and activity in vitro.

HPLC method development
Co-injected with Epitalon to set separation, retention and peak-purity criteria for both forms.

Stability-indicating methods
Used in forced-degradation studies alongside Epitalon to distinguish terminal from internal degradation routes.

MS identity marker
Its +42 Da (acetyl) and −1 Da (amide) shifts relative to Epitalon make it a clear identity marker in MS workflows.

05

Stability & Handling

Storage
Store the lyophilized powder at −20 °C, sealed, desiccated and protected from light.

Reconstitution
Soluble in water; dilute acetic acid or an assay buffer can be used depending on the downstream experiment.

Asp-Gly isomerisation
As with Epitalon, succinimide and isoaspartate formation at the Asp-Gly bond is fastest at mildly acidic pH (around 4–6) and higher temperatures. Keep solutions near neutral pH, cold, and use them promptly.

Enzymatic breakdown
The caps slow but do not stop degradation, so characterise stability in the actual assay matrix rather than assuming it.

Solutions
Freeze single-use aliquots and avoid repeated freeze–thaw cycles.

06

Analytical & Purity Considerations

Analysis must confirm both terminal modifications and separate the analogue from Epitalon.

HPLC
Reversed-phase C18 with a 0.1% TFA gradient, starting at low organic content. With its terminal charges removed, it usually elutes later than Epitalon, and the two separate well. Detect at about 205–214 nm.

Mass spectrometry
Positive-mode ESI gives [M+H]⁺ at m/z 432.2 (monoisotopic 431.17 Da), compared with 391.1 for Epitalon. MS/MS of the intact peptide confirms the acetyl group on N-terminal fragments and the amide on C-terminal fragments.

Impurity profile
Typical checks include incompletely capped forms (non-acetylated, about −42 Da; free acid, about +1 Da), isoAsp variants, the succinimide intermediate (−18 Da) and counter-ion content.

Frequently Asked Questions

Is this the same as Epitalon?

No. It has the same Ala-Glu-Asp-Gly sequence, but with an acetyl group on the N-terminus and an amide at the C-terminus. That makes it a different compound: about 41 Da heavier (431.4 versus 390.4 g/mol), with no free terminal charges.

Is it intended for human or animal use?

No. It is supplied strictly as a reference compound for laboratory and analytical research and is not for human or animal use.

What is the purity?

The specification is ≥98% by HPLC. The measured purity for each lot is reported on its certificate of analysis.

What is the advantage of the capped analogue over Epitalon?

Capping both ends blocks aminopeptidases and carboxypeptidases, so the analogue is expected to resist enzymatic breakdown better in biological samples. Whether it behaves the same as Epitalon in a given assay should be checked directly.

How should it be stored once received?

Keep the sealed vial at −20 °C, desiccated and protected from light. After reconstitution, keep solutions near neutral pH and cold, use them promptly, or freeze single-use aliquots.

Does capping protect against all degradation?

No. The internal Asp-Gly bond can still form isoaspartate, and internal peptide bonds can still be cleaved by endopeptidases. Only the terminal routes are blocked.
