01
Survodutide (development code BI 456906) is a synthetic 29-residue peptide based on human glucagon that activates both the glucagon receptor and the GLP-1 receptor. It keeps glucagon’s length and much of its sequence, with substitutions that add GLP-1 receptor activity and improve stability.
A C18 fatty diacid is attached to Lys24 through a γ-glutamate and a short glycine–serine linker, enabling albumin binding. Published cell assays report sub-nanomolar potency at both receptors. Survodutide offers a glucagon-based route to GLP-1/glucagon dual agonism, in contrast to the oxyntomodulin-derived mazdutide. It is supplied as a lyophilized reference material for in vitro dual-receptor pharmacology and analytical work on lipidated peptides.
02
Survodutide stays close to glucagon in length and layout but changes key positions.
His1 and Ac4c2
The N-terminal histidine is kept. Position 2 is 1-aminocyclobutane-1-carboxylic acid (Ac4c), a cyclic, α,α-disubstituted amino acid that, like Aib in other analogues, blocks DPP-IV cleavage.
Glucagon-based core
Much of the glucagon sequence is retained for glucagon receptor activity, while substitutions such as Lys12, Glu16, Arg17, Ala18/19 and Asp21 tune the balance towards GLP-1 receptor activity and improve stability.
Lipidated Lys24
The side chain of Lys24 carries octadecanedioic acid (C18 diacid) linked through γ-glutamic acid and a Gly-Ser-Gly-Ser-Gly-Gly spacer. This seven-unit branch is sometimes described in databases as a second short chain, but it is a side chain on Lys24, not a cross-link between two peptide chains.
C-terminus
The chain ends in Trp-Leu-Glu-Ser-Ala-NH₂ with an amidated C-terminus. Glucagon’s methionine has been replaced, so the peptide contains no methionine.
Sensitive residues
Trp25 can photo-oxidise and Gln3 can deamidate. There is no asparagine, methionine or cysteine.
03
Survodutide is characterised in vitro at both target receptors.
Receptor potency
cAMP assays in CHO-K1 cells expressing the human receptors report EC₅₀ values of about 0.52 nM at the glucagon receptor and 0.33 nM at the GLP-1 receptor, indicating potent, relatively balanced dual activity.
Comparison with native hormones
In assays containing a small amount of plasma, published data describe GLP-1 receptor potency similar to native GLP-1 and glucagon receptor potency about six-fold lower than native glucagon. Because the lipid binds albumin, measured potency always depends on assay conditions.
Glucagon receptor readouts
In hepatocyte models, glucagon receptor engagement is studied through glucose output, glycogen breakdown and lipid metabolism, compared with GLP-1 receptor-only agonists.
Preclinical context
Animal studies have examined energy expenditure, food intake and liver lipid metabolism, including models of fatty liver disease. These are cited as background only.
Characterisation of the reference material itself is limited to in vitro and analytical work.
04
Dual GLP-1/glucagon reference
The glucagon-based co-agonist reference in panels with mazdutide, semaglutide and retatrutide.
Scaffold comparison
Paired with the oxyntomodulin-derived mazdutide to study how backbone origin and side-chain design affect receptor balance, chromatography and stability.
Branched side-chain analysis
Its peptide-based linker makes it a useful test case for LC-MS/MS methods that characterise lipidated side chains containing amino acid spacers.
Lipidated peptide HPLC
A test compound for reversed-phase methods for C18-acylated peptides, alongside semaglutide.

Storage
Store the lyophilized powder at −20 °C, sealed, desiccated and protected from light. Let the vial reach room temperature before opening.
Reconstitution
DMSO dissolves it well for concentrated stocks; use fresh, dry DMSO. Aqueous solubility depends on the salt form (for example trifluoroacetate or sodium) and on pH, so check the certificate of analysis or test a small amount first.
Light sensitivity
Trp25 can photo-oxidise. Use amber or foil-wrapped containers and handle under subdued light.
Deamidation
Gln3 can deamidate slowly at neutral to basic pH. Freeze reconstituted aliquots and minimise time at room temperature.
Surface adsorption
The lipid promotes sticking to plastics at low concentrations. Use low-binding polypropylene tubes and consider 0.1% BSA as a carrier in dilute working solutions.
06
Analysis must confirm the glucagon-based backbone, the Ac4c residue and the branched side chain on Lys24.
HPLC
Wide-pore (300 Å) C4 or C8 columns at elevated temperature usually give the best peak shape and recovery. The des-lipid peptide is much less retained and separates easily; exact conditions are set during method development.
Mass spectrometry
ESI-MS shows multiply charged ions that deconvolute to about 4231.6 Da, roughly 330 Da lighter than mazdutide. Use formic acid rather than TFA for LC-MS.
Side-chain confirmation
Digestion followed by LC-MS/MS of the fragment containing Lys24 confirms the attachment site and the full side chain: C18 diacid, γ-glutamate and the Gly-Ser-Gly-Ser-Gly-Gly spacer. Using more than one enzyme (for example trypsin with Glu-C or Asp-N) improves coverage, since trypsin does not cleave after the modified Lys24.
Impurity profile
Typical checks include the des-lipid peptide, side chains missing linker residues, Trp25 oxidation (+16, +32 Da), Gln3 deamidation (+1 Da), truncated sequences and the free-acid C-terminus.
07
Survodutide is a 29-residue glucagon-based peptide with Ac4c at position 2 and a C18 diacid on Lys24 via a glycine–serine linker. Mazdutide is a 34-residue oxyntomodulin-derived peptide with Aib at position 2 and a C20 diacid on Lys20 via γ-glutamate and two AEEA units. Their intact masses differ by about 330 Da.
Not in the sense of a cross-link between two chains. Lys24 carries a side chain made of a C18 diacid, γ-glutamate and a Gly-Ser-Gly-Ser-Gly-Gly spacer. Some databases describe this branch as a short second chain, but it is attached only through the lysine side chain. Analytically, the spacer’s amino acids give extra MS/MS fragments that help confirm the side chain.
The values of about 0.52 nM (glucagon receptor) and 0.33 nM (GLP-1 receptor) come from cAMP assays in CHO-K1 cells and show potent, relatively balanced activity. They depend on the cell system, receptor expression and especially albumin or serum content, so they are best compared with reference agonists run under the same conditions.
Published studies show activity at both receptors in cell assays, and animal studies, including models lacking one receptor, link its effects on energy expenditure, food intake and liver lipid metabolism to engagement of both the GLP-1 and glucagon receptors. These findings are background only.
Their peptide backbones and linkers differ, so they separate on wide-pore C4 or C8 reversed-phase columns with an optimised gradient. Mass spectrometry distinguishes them unambiguously (about 4231.6 Da versus 4113.6 Da).
Mainly Trp25, which can photo-oxidise, and Gln3, which can deamidate. Survodutide contains no methionine, asparagine or cysteine.
No. It is supplied strictly as a reference compound for laboratory and analytical research and is not for human or animal use.
